Instead, the prestigious event will see competitors cross the line on the outskirts of Jindera.
Race committee member Greg Featonby said with traditional finish point Lavington Hall now utilised as a COVID-19 testing centre, as well as traffic barriers installed in Lavington's Collins Street, it's made the finish line unworkable.
The race will still start in Wagga on Holbrook Road and will see riders travel through Mangoplah, Cookardinia, Morven, Culcairn and Walla before finishing on Urana Road in Jindera.
"We've decided to move it out of town, which unfortunately is a bit of a shame," Mr Featonby said.
"It's an iconic event that links the two cities, but it won't link the two cities anymore.
"It will still be the John Woodman Memorial Race, it just won't be from Wagga to Albury, it will be from Wagga to Jindera.
"Rider safety and public safety is the most paramount thing.
"Things have to change, but that's just the way it is."

Mr Featonby said the new finish spot is likely to remain in place for the future of the event.
It will also mean the track is 11 kilometres shorter, ultimately now impacting how race records are determined.
The event dates back to the 1940s and was cancelled for the first time last year since it was revived in 1986.
It had originally been postponed until November, before the call was made to pull the pin due to the COVID-19 pandemic.
Mr Featonby said the local event is one of the few point-to-point races in Australia and has attracted some big name competitors in years gone by.
"It's always attracted a very strong field," he said.
"If you go back through the honour rolls there's guys who have won Olympic gold medals and there's Australian champions," he said.
This year's race will take place on Sunday, April 11.
