Lions' coach Kodee Lowe said the home side is hoping to see plenty of support from the surrounding communities, with money raised from tin rattling at the game going towards the annual Good Friday Appeal.
While the two sides will go head-to-head, they'll also both have a shared goal.
"We want to give as much money to the kids as possible," Lowe said.
"Years gone by we've always rattled tins along the Wahgunyah side of the bridge, but this year we're just asking everyone to come down to the game and support that way.
"There's a fierce rivalry between the clubs and we normally double our gate for a normal home and away season game.
"To do it on a special day might even boost it up a bit higher."
He admitted both clubs have welcomed the opportunity with open arms.
"Around the Corowa-Rutherglen region there's always so many people in town over Easter along the river," Lowe said.
"I think it's the perfect time to come and watch some local footy."
It's set to be Cats' coach Steve Owen's first look at a game between the two clubs as he heads into his debut season as Rutherglen's playing coach.
ALSO IN SPORT:
Owen had signed to assist outgoing coach Daniel Athanitis for the abandoned 2020 season before later taking the reins.
"Hopefully if it goes well we can make it an annual thing," Owen said.
"The more people we can get to come along and support the Good Friday Appeal the better.
"We're just excited to see the season go ahead."
Rutherglen's Jim Hanrahan is set to make his debut at the club after spending the 2019 season in Cairns.
Wahgunyah will also have a number of new faces taking to the field, including Dean Ewing, Gavin Cathie, and Cooper Bennett.
Former local Adam Romey, who has most recently been playing in the Northern Territory, will also line-up in the side.
"Everyone's pumped and I think the sides are looking pretty level at the moment," Lowe said.
Wahgunyah has also planned a half-time Easter egg hunt for families at the game.
The opening round of the TDFL last weekend saw Barnawartha, Tallangatta, Kiewa-Sandy Creek and Yackandandah post their first wins of the season.
The seniors will commence at 2:15pm.
