Tiger forwards Jacob Conlan and Brayden O'Hara kicked six and five goals respectively in the 20.17 (137) to 6.16 (52) home victory.
"I reckon we had a really good pre-season, obviously getting Pat Scammell (two-time Olympic middle distance runner) in, he made us work and he's got the boys going well," O'Hara said.
The Roos had scored a confidence-boosting win over Wodonga in round one.
"Certainly after the game, the players realised, hang on, as much as we played well last week, we've still got a long way to go to match the top teams in the competition," first-year coach Peter German told Radio 2AY.
The Roos couldn't handle Fletcher Carroll (40 touches, two goals), co-coach Anthony Miles (38) and O'Hara (36), while Conlan bounced back after five behinds early against Wangaratta Rovers.
