Phil Davies, Andrew Urquhart, Tom Ellis, Tom Manifold, David Cato, Harry Chisholm, Noel Passalaqua and Jim Salter will travel by motorbikes and utes for three weeks through remote desert and station tracks.
It is the fourth trek undertaken by longtime friends known as the Monkey Mia Boys after their first adventure from Monkey Mia, Western Australia, to Byron Bay in 2018.
The expeditions have raised more than $125,000 for Shake It Up Australia Foundation, a not-for-profit organisation that promotes research into Parkinson's, a condition experienced by some group members.
Shake It Up Australia Foundation founder Clyde Campbell said the funds were earmarked for research using Induced Pluripotent Stem Cells, which allowed for personalised drug selection as everybody's Parkinson's was different.
"This 'intelligent medicine' has massive opportunities to target the right drugs for the right patients by using technology to our advantage," he said.
IN OTHER NEWS:
This month's journey will include Poeppel Corner, where the Queensland, South Australia and Northern Territory boundaries meet, the Hay River Track, Alice Springs, the Madigan Line and the Birdsville Track.
Noel Passalaqua's wife Kim said on Wednesday the travellers, presently out of mobile range, aimed to increase awareness of Parkinson's, stay active, test their limits and have fun.
"It's a great experience, especially as we all age," she said.
"They're still keeping that friendship group and being able to get out and do things together.
"If you slow down, age catches up with you and I think all of them are trying to make sure that doesn't happen."
