There are 89 exposure sites linked to the latest outbreak with the most concerning being a Flemington public housing tower.
Victorian Premier Daniel Andrews said the origins of the two clusters were still unclear, but all cases were linked.
The state is receiving almost 150,000 additional doses of Pfizer following talks with the federal government.
"This Delta variant spreads so fast," Mr Andrews said.
"This will go where the vaccine isn't.
"The only options we've got is these lockdowns.
"This is a very concerning day."
There remains no cases in regional Victoria and no incursions from NSW confirmed in the latest outbreak to date.
Positive wastewater detects in Benalla have been reported in recent days.
In reference to Wangaratta reports of wasteawater detects, Mr Andrews said: "We apologise" and "nothing's perfect".
UPDATED: NSW recorded 319 new cases of COVID-19 in the 24 hours to 8pm on Friday night.
NSW Health has been notified of five more deaths from coronavirus with 28 total deaths linked to this outbreak.
Health Minister Brad Hazzard was concerned COVID-19 was seeping out of the eight local government areas in Greater Sydney under hard lockdowns and numbers could climb even higher in coming days.
He said Greater Sydney had the toughest lockdown measures in the country presently, but people weren't complying with the rules.
There are still no plans for a ring of steel around Sydney.
"I still harbour hope we will get back to zero (cases)," Mr Hazzard said.
"If we don't get the vaccine and don't follow the rules we won't get back to zero."
Almost 200 cases in NSW are still under investigation.
Armidale in northern NSW is going into lockdown from 5pm Saturday and last seven days.
There are two confirmed cases in the area with a sewerage detection of COVID-19 at Dubbo.
Newcastle was put into lockdown on Friday.
NSW Premier Gladys Berejiklian didn't attend the daily briefing on the day her state cracked 300-plus cases.
EARLIER: Victoria has recorded 29 fresh cases of COVID-19 in the past 24 hours with strong indications a hard border closure is returning to Albury-Wodonga.
The state's contact tracers are scrambling to get on top of the latest outbreak in Melbourne that has plunged Victoria into a sixth lockdown.
Infrastructure for a tighter border closure was being moved into place on the Hume Freeway late on Friday night.
All fresh cases are linked to previously reported ones, but were not in isolation during their infectious period.
