Van Berlo visited a specialist in Melbourne on Tuesday who confirmed he'll miss at least three weeks.
He has had a long-running injury history, including a reoccurring hamstring complaint that started in 2014.
It was already a delayed start to the season for Van Berlo who was suspended for two matches for rough conduct against Albury's Mathew Walker in last year's grand final.
Van Berlo "nicked" his quad in the second quarter of the Pies' round three victory against North Albury - his first appearance of the season.
Wangaratta coach Luke Morgan revealed Van Berlo won't be available before the interleague bye on May 18.
"He's got a nick in the quad and will have a bit of time on the sidelines," Morgan said.
"We've had a number of specialist appointments for him and he seen one yesterday (Tuesday in Melbourne).
"He's got a little bit of work to do and hopefully it recovers okay.
"Injury has plagued him.
"Hopefully he's back sooner rather than later."
Other sports news:
It's a major setback for Wangaratta ahead of its top-of-the-table clash with Lavington at Norm Minns Oval on Saturday, but Morgan said it provides an opportunity for someone else to step up.
"We're really looking forward to it," he added.
"We had a great session (on Tuesday night) and will come up against a team that's in really good form and playing some really good footy.
"We've had a productive start and probably achieved what we've wanted to achieve in the first four games, but we know we've got some more improvements to work on.
"These next two weeks against probably the premier teams (Lavington and Albury) in the competition will be exciting and a great challenge for our group."
Wangaratta has 11 players in the Ovens and Murray interleague training squad.
