The incident occurred just after 7am near the Great Alpine Road overpass.
Initial reports on social media suggest a truck and a vehicle collided in the southbound lanes and the truck has rolled.
An Ambulance Victoria spokesman said a man in his 30s was airlifted to the Royal Melbourne Hospital in a stable condition.
The cause of the crash was unclear.
About 8.15am, both southbound lanes were closed and the truck remained on its side.
A traffic detour of about 200 metres was in place, with vehicles leaving the freeway briefly at the Great Alpine Road exit.
The Department of Transport issued a traffic alert at 8.19am about the freeway closure.
"Motorists are advised to detour at the Great Alpine Road exit to get back on to the Hume Freeway," the alert said.
"Northbound lanes remain open."
The Victorian Department of Transport states that the Hume Freeway is "experiencing higher traffic volumes with the Princes Highway closed between Cann River and the New South Wales border due to bushfires".
