Ambulance Victoria have confirmed two people have died during a train derailment near Wallan, along the Albury line.
It's believed the incident happened near the Hume Highway.
The XPT was headed to Southern Cross from Sydney.
CFA said five passenger carriages derailed. A triage centre has been set up at the BP Service Station.
An Ambulance Victoria spokesperson said one person was being airlifted to hospital and a large number of people were being assessed but not believed to be seriously injured.
They said four people will be taken to hospital in a stable condition while a number of other people will be taken to hospital with minor injuries.
About 160 passengers were on board.
Emergency services were called at about 7.45pm.
It is believed the train left Sydney at 7.40am and was running more than two ours late at the time of the crash.
Passengers expecting to take the XPT from Melbourne to Sydney tomorrow, have been told 'due to an operational issue' the service will be replaced by coaches.

Police, Ambulance Victoria, State Emergency Service, CFA and air ambulances are responding to reports of a train derailment near Wallan.
Train services along the Albury and Seymour lines have been stopped.
IN OTHER NEWS:
At this stage it is unclear how many people have injured in the derailment or whether any fatalities have occurred.

On Twitter, V/Line told passengers services along the Shepparton and Seymour line were terminating early while a NSW Train Link investigation occurred near Wallan.
Earlier, V/Line told passengers to factor in an additional 60 minutes for trips after a signal hut at Wallan was destroyed by fire earlier this month.
