VicRoads says all Sydney-bound traffic is being diverted by emergency services onto the Benalla exit.
Traffic is then able to re-join the Hume Freeway via Sydney Road.
Heavy vehicles should use Mansfield Road, Goodwin Street, Ackerly Street, Midland Highway, Link Road, Benalla-Yarrawonga Road and Sydney Road back onto the Hume Freeway.
Melbourne-bound traffic is unaffected.
This closure is under Victoria Police control. The freeway will re-open to northbound traffic when investigations are complete and it's safe to do so.
"Our signals team is monitoring conditions through Benalla and will adjust traffic lights as needed to minimise congestion in the area," VicRoads said.
