Superintendent Paul Smith said there was a disproportionate amount of road trauma in rural and regional areas.
"That's very unfortunate," he said.
"That's our friends or family that are on those roads, sharing it with people who may be impaired by alcohol or drugs, or may be speeding.
"That's why our police are out there enforcing these laws and making the roads safer."
Highway patrol and general duty officers are targeting speeding, drink and drug driving, and other offences from Friday to Sunday as part of Operation Chrome.
"There will be significantly more police, from here to Moulamein, right across the district," Superintendent Smith said.
"Some people are thinking because of COVID-10, it's fine to get about, have a drink or two and the police won't stop you.
"That's not the case."

Coronavirus restrictions were eased in NSW on Friday, but Superintendent Smith said the rules still applied.
Officers continue to investigate recent crashes in Albury, including an incident where a stolen Mercedes SUV was dumped in the Murray River on Wednesday night or Thursday morning.
The vehicle he was in crashed into metal bars on a traffic island near the intersection of Fairview Drive about 5.15am Thursday.
