Resort staff checked cars on the outskirts of Mount Beauty and Harrietville in the first weekend with the rule.
Large electronic signage was erected in each township but some metropolitan visitors still arrived at checkpoints without having had a test in the previous 72 hours.
Falls Creek Coach Service owner Heath Fallon said the government requirement affected trade.
"It's slowed things down in comparison to last weekend," he said.
"We run up and down the hill about every two hours.
"Every day, we will send in a form with our passenger numbers to the resort and the drivers are checking for test results if people are from Melbourne.
"It seems to be running alright so far - I would definitely prefer not to do it."
While the state government has not indicated how long resorts will be required to enforce the test rule for Melbourne visitors, Mr Fallon hoped it would be lifted before the school holidays.
"Logistically, that would be really difficult - there would be a line right back to Mount Beauty," he said.
"We're not authorised officers ... but as long as we show our due diligence, we're doing what we can."
The test requirement did not result in any cancellations for Snowgum Motel in Mount Beauty.
"We have been telling people about it. It's been pretty comparable to most early ski seasons," manager Darren Tomkins said.
"We're pretty much full for the school holidays and people are forward-booking; all we need is the snow."
Mr Tomkins assumed the test requirement would last the season and said if it kept the resort open, so be it.
"We don't want to get shut down again," he said.
"Especially for the guys up on the mountain; they only have a short season to make their money."
The checkpoints provided an early opportunity for ensuring compliance of snow-chain-carrying.
Joseph Paterson of Hoys Harrietville Ski Centre said his team were reminding customers of the checks.
"It's great to have them back - most people so far have been very understanding," he said.
"We were still quite busy last weekend with regional Vic.
"We've been giving everyone the heads-up, letting them know they will need their test.
"There are a few disgruntled people ... but everyone gets that it's to protect the season."
