As one of five friendly games between the clubs, on a weekend when AWFA has cancelled all competitive matches, the annual senior men's fixture will still go ahead at Aloysius Park, kick-off 2:50pm.
Bill Luders and Hans Laramee were part of the steering committee that formed the Albury-Wodonga Soccer Association in 1973.
"Both clubs have agreed to play for the cup even though we're not playing for AWFA points," Hotspurs president Brad Howard said.
ALSO IN SPORT:
"We've got to take whatever we can get at the moment. Everyone just wants to play the game.
"It's something people want back in their lives, a little bit of normality."
Hotspurs and Melrose's junior boys will go head-to-head at under-12, under-15 and under-17 level and the clubs will also play a Division 2 women's match.
In addition, Hotspurs will take on Albury United in a Division 3 men's game.
