The business, which operated out of Daintree Medical Centre for three years, has moved to level two of the Wodonga Place Woolworths centre.
Principal Michael Bowler said the large gym space overlooking Junction Place was an attraction in deciding on their new space.
"We wanted to be closer to town, and to have a gym facility where people could become more active in their own rehab," he said.
"Part of what we are trying to do is get some consistency across all our clinics."
Michael and Simone Bowler took over Healthfocus Physiotherapy in 2018, with Mr Bowler staring with the business in 2004.
The new Wodonga site with seven treatment rooms is of a similar size to the Central Albury clinic and there is also a Lavington site.
Business manager Emily Shanahan said the first clients were through the doors on Monday, after many months of planning.
"We started on this earlier in the year, COVID hit and we put it on hold for a little bit," she said.
"We didn't have to close the doors ... things did slow down during the first wave, but business is starting to pick up as confidence is growing.
"We picked it back up mid-year, working with Connelly Construction, and they finished the build off for us in about 12 weeks.
"We've stayed local with all our trades, to get some money back into the local economy."
The opening of Healthfocus Physiotherapy comes after Squad, formerly ATEL, also opened a new office in the complex.
Ms Shanahan said Healthfocus had received a warm welcome from Squad and was excited about being part of the rejuvenation of Wodonga's CBD.
"I think the Wodonga Council has done an amazing job with central Wodonga and we're really excited to see the other developments," she said.
"It's a good, central spot; we're nice and close to everything and people can come see their physio and then grab a coffee or some lunch.
"It's been really positive, we're loving the space.

"We haven't had a gym space in our Wodonga clinic for a few years now, so it's nice to get back to that."
Riverina Podiatry Group would be co-sharing the space on Wednesdays and Fridays, with plans for group classes including Pilates.
"We are doing a new class, a spinal rehabilitation class, on Tuesday mornings, which we haven't offered in Wodonga for a few years," Ms Shanahan said.
